Mae ein labordy yn defnyddio modelau vivo ynghyd â samplau clinigol i ddehongli mecanweithiau moleciwlaidd croen a achosir gan farwolaeth celloedd a llid ar y cyd.
Necrosis wedi'i raglennu a larymau mewn llid
Ystyriwyd yn hir fel ffurf oddefol o farwolaeth celloedd, yn ddiweddar dangoswyd bod necrosis yn broses reoleiddiedig, wedi'i actifadu gan amrywiaeth o ysgogiadau mewndarddol neu alldarddol, Derbynyddion Marwolaeth o'r fath (DRs), ymyrryd (IFNs), neu Derbynnydd tebyg i Doll (TLRs). Mae marwolaeth Necrotig yn hynod o lidiol oherwydd rhyddhau moleciwlau mewngellog, o'r enw larymau neu Batrymau Moleciwlaidd Cysylltiedig â Pherygl (DAMPs). Necroptosis yw'r ffurf nodwedd orau o necrosis wedi'i raglennu a dangoswyd ei fod yn achosi llid mewn nifer o organau, yn enwedig mewn epithelia rhwystr (croen, colon, aren). Mae'n cael ei reoleiddio gan y cymhleth necrosome sy'n cynnwys kinases RIPK1 a RIPK3 a'u swbstrad, pseudo-kinase MLKL.
Gan ddefnyddio dulliau genetig ac mewn modelau vivo, nod ein gwaith yw egluro rôl necrosis a larymau wedi'u rhaglennu, yn ogystal â dehongli'r rheoliad moleciwlaidd o necrosis wedi'i raglennu mewn clefydau llidiol, mewn cydweithrediad â chlinigwyr, i nodi biofarcwyr cynnar a thargedau therapiwtig newydd ar gyfer meddygaeth wedi'i phersonoli.
