Mae fy ymchwil yn canolbwyntio ar y mecanweithiau cellog a rhwydwaith sy'n gweithredu yn y thalamws a'r cortex yn ystod epilepsi cwsg ac absenoldeb, gan ganolbwyntio ar gynulliadau niwronol ac astrocytig. Mae fy grŵp amlddisgyblaethol yn defnyddio cyfuniad o dechnegau microsgopig sganio laser electroffisiolegol, morffolegol, imiwnocemegol, cyfrifiadol a 2-photon mewn anifeiliaid arferol a modelau trawsgenig. Mae'r technegau hyn yn cael eu cymhwyso in vitro (diwylliant sylfaenol, sleisys ymennydd), yn ogystal ag mewn paratoadau anaestheteiddio ac yn ymddwyn yn rhydd. Rydym hefyd yn defnyddio sbectrosgopeg MRI i fesur niwrodrosglwyddyddion a'u metabolion yn ymennydd plant a phobl ifanc ag epilepsi absennol.

Ymchwil
Mae fy ymchwil yn canolbwyntio ar y mecanweithiau cellog a rhwydwaith sy'n gweithredu yn y thalamws a'r cortex yn ystod epilepsi cwsg ac absenoldeb.
Yn ddiweddar, fe wnaethom hefyd ddatblygu diddordeb yn y mecanweithiau sy'n ymwneud â signalau astrocyte-niwron.
Mae fy ngrŵp amlddisgyblaethol yn defnyddio cyfuniad o dechnegau delweddu electroffisiolegol, morffolegol, imiwnocemegol a chonfocal mewn modelau arferol a thrawsgenig mewn vivo ac mewn modelau anifeiliaid in vitro, yn ogystal â dull cyfrifiadurol ar gyfer efelychu gweithgareddau rhwydwaith niwronol ac astrocytig.
Mecanweithiau cysgu
Un o'r gweithgareddau trydanol mwyaf sylfaenol sy'n digwydd mewn niwronau thalamig a chortigol yn ystod cwsg naturiol yw'r 'osgiliad cwsg araf'. Mae'r panel uchaf yn Ffigur 1 isod yn dangos enghraifft nodweddiadol o'r gweithgaredd hwn a gofnodwyd o niwron thalamig.
Mae'r 'osgiliad cwsg araf' yn cael ei nodweddu gan bresenoldeb dau botensial bilen (a elwir yn wladwriaethau UP a DOWN), sy'n cael eu cynhyrchu trwy newid 'ymlaen' ac 'i ffwrdd' cydrannau ffenestr y foltedd isel a weithredir, T-math Ca2+ cyfredol (I T). Yn y panel gwaelod o Ffigur 1, mae'r ddau gerrynt niwronal eraill sy'n chwarae rhan hanfodol yn yr osgiliadau araf yn cael eu darlunio, hy Ydw, wedi'i actifadu gan hyperpolarization Na + / K + cyfredol, ac I CAN, Ca2 +-activated cerrynt cation detholus. Mecanweithiau pathoffisiolegol Epilepsi Absenoldeb
Mae epilepsi absenoldeb yn fath anorchfygol o epilepsi sy'n effeithio ar blant a phobl ifanc yn bennaf. Mae pob trawiad yn cynnwys nam sydyn a byr ar ymwybyddiaeth, sy'n cyd-fynd â syllu gwag, diffyg ymateb i symbyliadau allanol, ac ymddangosiad gweithgaredd patholegol nodweddiadol, a elwir yn 'ryddhad spike-and-wave' (dangosir yn Ffigur 2A, dde).
Mae'r 'rhyddhau sbectol' yn cael ei gynhyrchu gan weithgareddau trydanol annormal niwronau cortigol a thalamig (a ddangosir yn Ffigur 2B-D, mae cod lliw yn cyfeirio at y gwahanol fathau o gelloedd a ddangosir yn y diagram sgematig). Sylwch ar danio'r niwronau GABAergic cryf iawn yn y niwclews reticularis thalami (NRT) (gweler y ffilm isod) a thawelwch trydanol y niwronau thalamocortical (TC).
Gan mai niwronau TC yw'r rhai sy'n trosglwyddo symbyliadau synhwyraidd, mae eu hyperpolareiddio a'u diffyg tanio yn egluro pam nad yw plant yn ymatebol yn ystod trawiad ar absenoldeb.

signalau astrocyte-niwron
Nid yw astrocytes (math o gelloedd glial) bellach yn cael eu hystyried fel darparu cefnogaeth fecanyddol yn unig i niwronau ac yn rheoli crynodiad allgellog ïonau a metabolion allweddol, ond dangoswyd eu bod yn cyfrannu'n weithredol at drosglwyddo gwybodaeth niwronol ar y lefel synaptig.
Gan fod astrocytes yn gelloedd na ellir eu cyffroi mae eu 'actifadu' yn cynnwys codiadau dros dro mewn Ca2+ mewngellog sydd wedyn yn arwain at ryddhau trosglwyddyddion yn fetrigl, gan gynnwys glutamad ac ATP.
Mae'r sylweddau hyn a ryddhawyd yn seryddol wedyn yn gweithredu ar niwronau (gan actifadu derbynyddion NMDA yn ddelfrydol) rhyddhau trosglwyddydd modiwleiddio neu reoli effeithiolrwydd synaptig. Yn wir, gall astrocytes hyd yn oed gynhyrchu tonnau Ca2 + digymell a lleol, yn absenoldeb unrhyw weithgaredd niwronau.

Mae gan fy labordy chwe gorsaf electroffisiolegol (tair ar gyfer recordiadau patsh- a thri ar gyfer recordiadau miniog-electrod), un microsgop confocal (Odyssey, Thermo Noran, UDA) gyda chyfleusterau recordio patch-electrod in vitro , un microsgop sganio laser 2-photon (Ultima, Prairie Technology, UDA) gyda chyfleusterau recordio vivo ac in vitro patch- a sharp-electrode, cyfres ar gyfer in vivo Recordiadau trydanol a chymhwyso cyffuriau lleol mewn modelau sy'n symud yn rhydd, un clwstwr o nodau prosesydd deuol 11 ar gyfer efelychiadau cyfrifiadurol, meddalwedd a ddatblygwyd yn fewnol ar gyfer arbrofion clamp deinamig, a chyfleusterau ar gyfer dadansoddiad morffolegol ac imiwnocemegol ôl-hoc o niwronau a astrocytes.

Addysgu
Yn Ysgol y Biowyddorau ym Mhrifysgol Caerdydd, rwy'n addysgu niwrowyddoniaeth a niwroffisioleg i fyfyrwyr gwyddoniaeth israddedig (modiwlau Bi2432, BI3451, Bi3452) ac i fyfyrwyr deintyddol. Yn y Gyfadran Meddygaeth a Llawfeddygaeth ym Mhrifysgol Malta, rwy'n addysgu niwrowyddoniaeth a chlefydau niwrolegol i fyfyrwyr meddygol.
Rwyf hefyd yn goruchwylio myfyrwyr Meistr a PhD yn y ddwy Brifysgol.
