Ymchwil
Metaboledd a Swyddogaeth Lipidau Acyl
Straen amgylcheddol a metaboledd lipid
Mae ymchwil yn cynnwys ystod eang o ffactorau amgylcheddol ac organebau targed yn amrywio o ficrobau i blanhigion uwch.
Canfuwyd bod yr Effaith Tŷ Gwydr (CO2 a thymheredd atmosfferig uwch) yn newid metaboledd lipid, yn unol â'i weithred wrth ysgogi biogenesis bilen a thwf. Gellir esbonio'r newidiadau mewn metaboledd trwy fynegiant newidiol o ddau ensym allweddol sydd bellach yn cael eu profi'n uniongyrchol. Yn gysylltiedig â'r gwaith hwn mae arbrofion ar addasu tymheredd yn y protozoon pridd pwysig, Acathamoeba castellanii. Mae effeithiau tymheredd isel wedi'u gwahanu oddi wrth effeithiau ocsigen wrth gymell mynegiant o ddirwasgiad asid brasterog n-6 sy'n gwneud y lipidau bilen yn fwy 'hylif' ac yn caniatáu ffagocytosis, a fyddai fel arall yn stopio ar dymheredd isel.
Mae rheoleiddwyr twf planhigion, fel auxins, yn ysgogi biosynthesis lipid bilen mewn pys ac rydym wedi astudio rheolaeth ffurfio ffosffatidylcholine. Mae pwynt rheoli pwysig ar lefel cytidylyltransferase ac rydym wedi astudio ei reoleiddio yn ogystal ag ynysu ei genyn. Yn fwy diweddar, rydym wedi ynysu'r genyn ar gyfer colin kinase ac wedi puro ac astudio'r ensym. Cyfansoddyn bilen planhigion pwysig arall yw'r sylffolipid. Rydym wedi egluro ei lwybr biosynthetig a'i ddiraddiad. Mae'r olaf yn bwysig ar gyfer ail-feicio sylffwr sy'n gorfod digwydd bob hydref (Fall) wrth i ddail gael eu diraddio. Mae'r broses yn cynnwys llwybr ffug-glycolytig gan ddefnyddio bacteria pridd.
Mae sawl dosbarth o blaladdwyr yn gweithredu trwy eu dylanwad ar metaboledd lipid. Rydym wedi astudio sawl ffwng sy'n lladd ffyngau planhigion (malltod tatws, llwydni) ac sy'n ymddangos fel pe baent yn newid metaboledd lipid. Mae astudiaethau hirsefydlog hefyd yn ymwneud â thiocarbamates (dosbarth chwynladdwr sy'n effeithiol trwy atal elongation asid brasterog ac, felly, ffurfiant haen wyneb) a graminicides.
Mae diddordeb cynyddol yn effeithiau gwenwynig metelau trwm. Rydym wedi dangos bod metelau o'r fath yn ymyrryd â metaboledd lipid ar wahanol lefelau mewn algâu morol, bryoffytau a chennau. Mae rhai bryoffytau braidd yn gwrthsefyll her plwm neu gopr a gellir eu datblygu ar gyfer bioadfer. O algâu brown, rydym wedi gallu ynysu metallothionein sy'n bwysig ar gyfer homeostasis metel arferol.
Rheoleiddio synthesis lipid mewn cnydau olew
Mae cnydau olew yn cynhyrchu tua 135 miliwn tunnell o gynnyrch, sy'n werth tua 115 biliwn US $ bob blwyddyn. Mewn termau meintiol palmwydd olew yw'r pwysicaf (cyfanswm o tua 25%), ffa soia yr ail (cyfanswm o 22%) a threisio hadau olew y trydydd (cyfanswm o 13% o olew). Mae angen amlwg i gynhyrchu mwy o olew, nid yn unig ar gyfer defnyddiau bwytadwy ond hefyd fel porthiant cemegol adnewyddadwy. Ar ben hynny, mae deall sut mae ansawdd yn cael ei reoli hefyd yn bwysig. Ni oedd y labordy cyntaf i gymhwyso'r dechneg o ddadansoddi rheoli fflwcs i synthesis lipid ac ers hynny rydym wedi ei ddefnyddio i archwilio rheolaeth fetabolig mewn cnydau pwysig fel palmwydd olew, ffa soia, had trais rhywiol ac olewydd.
Mewn palmwydd olew mae'r prif reolaeth yn cael ei weithredu gan gyflenwi asidau brasterog. Felly, dylid canolbwyntio ymdrechion i gynyddu cynnyrch olew mewn llinellau palmwydd olew sydd â chyfraddau uwch o synthesis asid brasterog. Er bod palmwydd olew 5 - 10 gwaith mor gynhyrchiol â chnydau olew eraill (fesul hectar), mae potensial o hyd i gynyddu ei gynnyrch 2 - 3 plygu. Byddai hyn o fudd mawr i'r byd.
Brassica napus (rape hadau olew) yw'r prif gnwd olew yng Ngogledd Ewrop a Chanada. Mae'r rhan fwyaf o blanhigfeydd yn defnyddio'r mathau isel o ddŵr sy'n addas ar gyfer olewau bwytadwy. Wrth astudio rheoleiddio cronni olew yn B. napus , gwnaethom ragweld y byddai llinellau â diacylglycerol acyltransferase uwch (DGAT) yn cynhyrchu mwy o olew. Dangoswyd bod llinellau trawsgenig gyda gweithgaredd DGAT uchel yn cynhyrchu 8% o gynnyrch cynyddol yn gyson ac yn fwy cadarn mewn amodau sychder. Rydym nawr yn edrych i weld sut y gellir cael cynnydd pellach mewn cynnyrch.
Biocemeg lipid ac agweddau meddygol
Mae nifer o brosiectau yn y labordy. yn archwilio rôl lipidau acyl mewn problemau meddygol.
Mae ysgytwad yr ysgyfaint, sy'n cael ei secretu'n barhaus i'r alfeoli, yn atal cwymp yr ysgyfaint ar anadlu allan (diwedd dod i ben). Nid yw babanod cynamserol yn gwneud syrffactydd ac, felly, gallant ddioddef o drallod anadlol. Roeddem yn un o'r labordai cyntaf i ddylunio a phrofi syrffactydd artiffisial – mae cymysgeddau o'r fath mor llwyddiannus nes eu bod yn cael eu defnyddio'n rheolaidd y dyddiau hyn. Aethom ymlaen i ddangos sut y gall metaboledd syrffactydd gael ei ddylanwadu'n sylweddol gan lwch (e.e. silica) anadlu a pin pwyntiodd y mecanwaith moleciwlaidd. Mae secretion tebyg sy'n llawn ffosffolipidau hefyd yn cael ei ffurfio yn y ceudod peritoneal lle mae ei effeithlonrwydd yn gysylltiedig â phroblemau gyda dialysis arennol ac adweithiau llidiol sy'n gysylltiedig â adlyniadau llawfeddygol.
Mae llid hefyd yn chwarae rhan mewn sioc endotoxic — adwaith acíwt gan rai cleifion i heintiau bacteriol negyddol gram yn dilyn llawdriniaeth. Rydym wedi dod o hyd i gydberthynas rhwng metaboledd asidau brasterog amlannirlawn mewn lipidau pilen penodol a rhagddodiad i sioc endotoxic. Dilynwyd y darganfyddiad hwn gan nodi acyltransferases penodol a all gyfryngu'r newidiadau mewn metaboledd. Astudiwyd un o'r ensymau hyn yn fanwl a nodwyd ei genyn. Yn ddiddorol, mae'n dangos newidiadau mewn lleoleiddio celloedd yn ystod ysgogiad cytokine.
Mae arthritis yn glefyd eang sy'n achosi llawer o ddioddefaint ac sy'n cael ei gyfryngu trwy lid gormodol a chronig. Rydym wedi bod yn astudio'r mecanweithiau moleciwlaidd y tu ôl i'r rhyddhad a ddarperir gan ddeietau olew pysgod sy'n gyfoethog mewn asidau brasterog amlannirlawn n-3 (PUFA). Gall asidau o'r fath (ond nid mathau eraill o asidau annirlawn neu dirlawn) leihau mynegiant o cytocinau llidiol ac eicosanoidau a gweithgaredd cartilag - gan ddiraddio proteinases mewn diwylliannau chondrocyte gwartheg. Yn ogystal, rydym yn treiddio'r llwybrau signalau sy'n sail i'r effeithiau ar drawsgrifio. Rydym wedi ymestyn y canlyniadau cyffrous hyn i dreialon clinigol yn ogystal ag arddangosiadau bod PUFAs dietegol n-3 yn effeithiol iawn mewn bwyd cŵn, yn enwedig ar gyfer bridiau sy'n dueddol o arthritis. Maent bellach yn cael eu defnyddio mewn llawer o frandiau perchnogol.
Mae clefydau eraill sydd â llid cronig fel ffactor sylfaenol pwysig yn cynnwys clefyd cardiofasgwlaidd (CVD) a dementia. Ar gyfer CVD, gellir dangos bod PUFAs n-3 o fudd i unigolion sydd mewn perygl o glefyd y galon. Rydym wedi dangos eu bod yn lleihau'r defnydd o lipoprotein dwysedd isel gan macrophages ac, felly, gallent leihau ffurfiant celloedd ewyn. PUFA arall (dietegol) o fudd posibl yw asid dihomogama-linolenig (DGLA) sydd, er bod pufa n-6, yn cael ei drawsnewid yn gynhyrchion gwrthlidiol ac yn lleihau llid mewn macroffagau.
Gyda disgwyliad oes cynyddol, mae dementia yn dod yn fwy a mwy cyffredin. Mae clefyd Alzheimer (AD) yn cyfrif am oddeutu 70% o gyfanswm yr achosion o ddementia ac mae'n costio dros £25 biliwn y flwyddyn i'r DU. Gellir lleihau'r llid sy'n gysylltiedig ag AD yn sylweddol gan ymyrraeth ddeietegol (olew pysgod neu pufa n-3). Mae hyn yn arwain at lai o ddyddodion beta-amyloid ac ymddygiad gwybyddol gwell. Yn ddiddorol, mae'r docosahexaenate ymennydd cynyddol (DHA) yn ddetholus ar gyfer y ffosffoglyceridau ethanolamine sydd wedi'u cysylltu ag AD.

Dros 640 o gyhoeddiadau gwyddonol gan gynnwys 5 llyfr a 14 o gyfrolau wedi'u golygu. Mae'r llyfrau yn cynnwys Lipidau: biocemeg, biotechnoleg ac iechyd ( Gurr, M.I. et al., chweched argraffiad, 2016, Wiley-Blackwell) sef y testun myfyrwyr datblygedig diffiniol ar lipidau. Rwyf wedi bod yn gyd-awdur ar y tri argraffiad diwethaf. Golygais Handbook of Olive Oil ( R. Aparicio and J.Harwood, ail argraffiad, 2013, Springer ) sydd wedi cael dau argraffiad a chyfieithiad i'r Sbaeneg. Dyma'r prif gyfrol gyfeirio awdurdodol ar olew olewydd. Rwyf hefyd wedi bod yn gyd-olygydd pob un o'r tri argraffiad o The Lipid Handbook, y prif waith cyfeirio ar lipidau ( Gunstone, FD, Harwood, J.L. a Dijkstra, A.J., eds.,trydydd argraffiad, 2007, Gwasg CRC ).
