Rwy'n uwch ddarlithydd mewn Bioleg Strwythurol. Rwy'n arbenigo mewn astudio siapiau moleciwlau biolegol ar y lefel atomig, i gasglu ffyrdd a mecanweithiau rhyngweithio rhwng gwahanol foleciwlau bywyd. Rwyf wedi cynnal ymchwil mewn llawer o wahanol feysydd Gwyddorau Bywyd, gan roi dealltwriaeth o gyfathrebu biolegol mewn amrywiaeth o systemau. Agorodd hyn y gallu i ddylunio therapïau ar gyfer gwahanol gwynion, maes wrth wraidd fy ymchwil. Mae hefyd yn gyfle i ddylunio diagnosteg ac i ddefnyddio biotechnoleg i alluogi prosesau nad ydynt yn digwydd yn naturiol.
Rwy'n raddedig mewn Cemeg, ar lefel BSc a PhD. Rwy'n defnyddio cymwysiadau Ffiseg i astudio macromoleciwlau biolegol. Mae gen i brofiad helaeth gyda crisialog pelydr-X a phenderfynu strwythur 3D gan ddefnyddio ymbelydredd syncrotron. Rwyf wedi defnyddio'r technegau cysylltiedig o Small Angle X-ray Scattering (SAXS) a Circular Dichroism (CD) i daflu goleuni ar fecanweithiau biolegol.
Rwyf wedi gwasanaethu ar bwyllgorau a chymryd rhan mewn rhaglenni ar lefel genedlaethol a rhyngwladol. Rwyf wedi cynnal llawer o ymarferion allgymorth a chyfryngau yn ystod fy ngyrfa, gan gyfathrebu perthnasedd uniongyrchol a phwysigrwydd Gwyddoniaeth mewn bywyd bob dydd.

Mae'r firws yn cael ei wneud yn gyntaf heb ei amlhau trwy gael gwared ar yr exons dyblygu, yna mae darn byr o niwcleotidau codio yn cael eu mewnosod ar bwynt strategol yn y protein targot ar sruface y firws i ddynwared rhan o'r protein sy'n gysylltiedig â chanser. Pan fydd y celloedd targed wedi'u heintio, mae'r epitop yn cael ei fynegi ac mae'r system imiwnedd yn cael ei symud i'w adnabod, gan ymosod ar bob cell malaen. Rydym wedi defnyddio penderfyniad strwythur yn helaeth i gael gwybodaeth am y strategaeth orau i'w defnyddio yn yr ymdrech hon, gyda thua 20 o gofnodion newydd i'r banc data protein. - Proteinau'r System Imiwnedd: Nod y gwaith hwn yw pennu'r ffactorau strwythurol sy'n gysylltiedig â chroes-adweithedd derbynyddion celloedd sy'n deillio o Thymus, TCRs, pan fyddant yn adnabod mwy nag un antigen a gyflwynir yng nghyd-destun y cymhleth Histocompatibility Mawr, mecanwaith sy'n sail i awtoimiwnedd. Ail agwedd yw dirywiad rhwymo TCR i wahanol antigenau a gyflwynir gan un MHC i ganiatáu cwmpas eang o antigenau posibl gyda'r nifer cyfyngedig o MHCs sydd gan unigolyn. Mae'r prosiect yn cynnwys defnyddio crisialog pelydr-X i ddatrys strwythurau 3D o'r endidau hyn, a chynnal astudiaethau rhwymo gyda chyseiniant plasmon wyneb, calorimetri titration isothermol ac actifadu celloedd T naïf. Cydweithio â'r Grŵp Modiwleiddio Celloedd T dan arweiniad yr Athro Andrew Sewell, yn y Sefydliad Heintiau ac Imiwnoleg, Prifysgol Caerdydd.
- Proteinau Fuorescent Gwyrdd Gwell: Mae'r protein fflwroleuol gwyrdd o Aequoris victoria yn cael ei ddefnyddio ar hyn o bryd mewn llawer o gymwysiadau biolegol a biotechnolegol i helpu i fapio dosbarthiad endidau biolegol mewn celloedd, trwy ei atodi i wrthgyrff a godwyd yn erbyn moleciwlau o ddiddordeb. Ar ôl rhwymo, mae'r GFP sy'n cario gwrthgyrff yn fflworoleuo ar donfeddi penodol. Nod rhaglen o beirianneg genetig y GFP o'r pysgodyn slefrod môr Aequoria victoria yw gwella ei briodweddau fflworoleuedd ar gyfer gwelededd gwell. - Tocsinau Pryfleiddiad Bacteriol Naturiol: Rydym yn astudio tocsinau naturiol amrywiol a gynhyrchir gan Bacillus thuringiensis ac organebau tebyg i ddeall eu dull gweithredu ac i ddatblygu plaladdwyr sy'n gyfeillgar i'r amgylchedd y gellir eu targedu at blâu penodol. Cydweithio â'r Athro Colin Berry yn yr Ysgol Bioscineces ym Mhrifysgol Caerdydd.
- Dyluniad Atalydd Calpain: Mae Calpain yn caspase aml-barth sy'n ymwneud ag ymateb i lid. Mae'n helpu celloedd gwaed gwyn i newid siâp i groesi trwy feinwe epithelial ger safleoedd llid. Mae gormod o weithgaredd yn gysylltiedig ag arthrhitis ar ôl lefel ymateb uwch. Roedd atal Parth VI wedi'i gyflawni yn y gorffennol, mewn mannau eraill, gan weithio gyda'r ensym moch. Yma, rydym yn gweithio ar yr ensym dynol, gan geisio optimeiddio'r atalydd ar gyfer affinedd uwch a gwell cydnabyddiaeth. Cydweithio â thîm dan arweiniad yr Athro Ruedi Allemann yn yr Ysgol Cemeg, Prifysgol Caerdydd.
- Biocoloration mewn cragen cimychiaid: Datryswyd strwythur β-crystacyanin, y protein lliw mewn cragen cimychiaid, gan ddefnyddio pelydr-X meddalach i ddata diffreithiant cam. Mae'r strwythur yn amlinellu'r mecanwaith lle mae golau yn cael ei anghyfreithloni a'i fodiwleiddio gan y protein i effeithio ar allyriad gwahanol yn y sbectrwm gweladwy trwy ddylanwadu ar gydffurfiad y cromoffor astaxanthin sy'n cael ei rwymo gan y protein. Datblygwyd methodoleg newydd i ddefnyddio'r signal anomalaidd o atomau S er mwyn datrys y strwythur. Cydweithrediad â'r Athro John Helliwell, Prifysgol Manceinion.
