Trosolwg o'r ymchwil
Mae homeostasis epithelial yn sylfaenol i iechyd a goroesi. Mae iechyd meinwe yn cael ei gynnal trwy brosesau sy'n rheoleiddio nifer a ffitrwydd celloedd mewn meinweoedd. Mae hyn yn sicrhau bod celloedd aberrant neu mutant - a fyddai fel arall yn achosi clefyd - yn cael eu tynnu. Mae cystadleuaeth celloedd yn disgrifio proses gyffredinol o 'rheoli ansawdd' sy'n arwain at ddileu celloedd 'coll'. Felly, dim ond celloedd o'r ansawdd uchaf sy'n cyfrannu at feinwe.
Mae ein hymchwil yn dangos bod cystadleuaeth celloedd yn digwydd mewn epithelia y pancreas oedolion ac mae hyn yn amddiffyn rhag canser. Ar hyn o bryd rydym yn ymchwilio i'r mecanweithiau sy'n pennu canlyniadau cystadleuaeth in vivo. Mae fy labordy wedi arloesi systemau celloedd a meinwe arloesol a dulliau rhyngddisgyblaethol i astudio cystadleuaeth celloedd mewn epithelia mamaliaid. Rydym hefyd wedi datblygu modelau llygoden newydd in vivo o ganser y pancreas a'r ysgyfaint digymell. Mae ein modelau arbrofol yn crynhoi datblygiad canser dynol yn fwy cywir oherwydd ein bod yn troi treigladau sy'n achosi canser mewn niferoedd isel o gelloedd. Mae'r dull hwn yn caniatáu inni ddiffinio'r bioleg celloedd moleciwlaidd sy'n sail i sut mae celloedd mutant genetig yn ehangu ac yn tyfu tra'n cael eu hamgylchynu gan gelloedd iach ac mewn meinwe 3D cymhleth. Rydym yn cyfuno modelau llygoden â delweddu meinwe 3D arloesol, bioleg celloedd meintiol, trawsgrifiad, gan gynnwys trawsgrifiad gofodol a bioleg gyfrifiadurol.
Cystadleuaeth celloedd mewn iechyd epithelial a chlefydau
Fe wnaethom nodi EPHA2 (derbynnydd tyrosine kinase o'r teulu Eph-ephrin o signalau dwyochrog) fel signal wedi'i gadw'n esblygiadol sy'n sbarduno diarddel celloedd mutant RAS o feinweoedd epithelial in vitro ac in vivo. Fe wnaethom ddangos yn ddiweddar fod signalau WNT5A hefyd yn rheoleiddiwr pwysig o dynged celloedd mutant RAS. Mae uwchreoleiddio signalau WNT5A yn caniatáu i gelloedd mutant RAS osgoi dileu celloedd a goroesi yn y meinweoedd.
Mae prosiectau cyfredol a ariennir gan BBSRC / UKRI yn ymchwilio a yw lleoliad meinwe yn pennu tynged celloedd mutant RAS ac a yw ffactorau auonomous ac anymreolaethol celloedd yn rhagweld tynged celloedd RAS.
Datgelu bioleg canser y pancreas cynnar
Mae canser y pancreas yn glefyd dinistriol oherwydd nad ydym yn gallu ei ganfod yn gynnar pan fyddai ymyrraeth glinigol yn gwella bywydau cleifion yn fawr. Mae canfod yn gynnar yn hanfodol i wella prognosis cleifion ac achub bywydau.
Mae tiwmorau pancreatig yn dechrau o gelloedd sy'n mynegi mwtaniadau KRAS oncogenig. Mae ein hymchwil yn dangos bod celloedd KrasG12D yn cael eu rhagori a'u dileu o feinweoedd pancreas iach in vivo ac mae hyn yn ataliol tiwmor. Fodd bynnag, mae mecanweithiau dileu yn aneffeithlon ac mae rhai celloedd KRAS yn osgoi dileu celloedd ac yn hytrach yn goroesi mewn meinweoedd fel celloedd segur a bôn-debyg. Ein nodau yw nodi ffactorau sy'n galluogi celloedd mutant RAS i osgoi dileu celloedd. Yn yr un modd, ein ffocws yw datgelu signalau / ffactorau sy'n hyrwyddo ehangu celloedd mutant RAS yn ystod camau cynnar canser y pancreas.

Lecturer in Biomedical Sciences, School of Biosciences since Sept 2020. I joined the European Cancer Stem Cell Research Institute (ECSCRI) as Research Fellow in 2013. Postdoctoral training in the laboratory of Dr Yasuyuki Fujita at MRC Laboratory for Molecular Cell Biology (LMCB), University College London. I carried out my PhD training in the laboratory of Professor Gareth Jones at the Randall Division of Cell and Molecular Biophysics, King's College London.
