Rwy'n Academydd Clinigol mewn Neffroleg ac yn Bennaeth Labordai Uned Ymchwil Aren Cymru yng Nghaerdydd. Mae gen i swydd Uwch-ddarlithydd Clinigol yn yr Ysgol Meddygaeth ym Mhrifysgol Caerdydd ac mae gennyf swydd Neffroleg Ymgynghorol er Anrhydedd ym Mwrdd Iechyd Caerdydd a'r Fro a Bwrdd Iechyd Aneurin Bevan.
Fy mhrif ddiddordebau ymchwil yw ym maes ffibrosis arennol. Nod fy grŵp yw deall a disgrifio'r mecanweithiau a all atal a/neu wrthdroi anaf acíwt a blaengar i'r arennau gan arwain at well iechyd yr arennau. Mae'r matrics allgellog yn chwarae rhan bwysig wrth gynnal iechyd biolegol yn ein celloedd a'n meinweoedd ac mae rheoleiddio matrics allgellog aberrant yn gyrru afiechyd, yn enwedig yn yr arennau. Mae fy ymchwil wedi dangos bod Hyaluronan yn glycosaminoglycans matrics allgellog, sy'n bwysig wrth reoleiddio ffenoteip celloedd stromal a swyddogaeth i ddylanwadu ar iechyd a chlefyd arennol. Mae fy astudiaethau'n canolbwyntio ar sut y gellir rheoleiddio'r gydran matrics hon i wella adferiad arennau yn dilyn anaf a chlefyd.
Mae fy meysydd ymchwil cyfochrog yn cynnwys ymchwil dialysis peritoneol (deall mecanweithiau sy'n gyrru ffibrosis yn y peritonewm) ac ymchwil cardiofasgwlaidd (deall gyrwyr cellog a moleciwlaidd mwy o batholeg cardiofasgwlaidd mewn cleifion â chlefyd yr arennau).
Mae gen i ddiddordeb sylweddol mewn addysg feddygol israddedig ac ôl-raddedig ac mae gen i swyddi addysgu arweiniol yn y ddau faes hyn. Roeddwn hefyd yn rhan o raglen SUSTAIN yr Academi Gwyddorau Meddygol pan gafodd ei sefydlu gyntaf yn 2015.

Maes Ymchwil Sylfaenol
Mae fy ymchwil wedi canolbwyntio ar fioleg celloedd a matrics fel y'i cymhwysir at glefyd arennol cynyddol. Mae gan 15% o boblogaeth y DU glefyd cronig yr arennau, gan eu rhoi mewn mwy o berygl o amlafiachedd a marwolaeth. Nid yw atal/gwrthdroi prosesau ffibr-amlochrog sylfaenol sy'n gyrru clefyd cronig yr arennau yn bosibl eto, ac mae ymarfer clinigol cyfredol yn rhagweld yn wael a fydd cleifion yn datblygu clefyd cronig yr arennau blaengar neu'n dangos adferiad arennol a/neu ddatrysiad. Nod fy ngrŵp yw nodi targedau moleciwlaidd a chellol newydd a all atal clefyd acíwt a chronig yr arennau i atal cleifion sydd angen therapïau amnewid arennol.
Mae Hyaluronan yn glycosaminoglycans matrics sy'n dangos mwy o fynegiant strymol ym mhob math o anaf acíwt a chronig i'r arennau, gyda'i fynegiant cynyddol yn cydberthyn â chanlyniadau arennol. Mae fy grŵp wedi nodi targedau newydd sy'n gysylltiedig â Hyaluronan a'i broteinau rhwymol a all atal a gwrthdroi ffibrosis mewn systemau celloedd vitro. Mae'r ymchwil gyfredol bellach yn canolbwyntio ar ddatblygu agweddau trosiadol ar yr ymchwil hon ac yn dangos perthnasedd a chymhwysedd y targedau cellog hyn i bennu eu defnydd fel biofarcwyr wrth ragfynegi dilyniant clefyd arennol, yn ogystal ag ymchwilio i'w potensial fel therapïau gwrth-fibrotig newydd.
Meysydd ymchwil cysylltiedig
Mae gen i ddiddordeb gweithredol hefyd mewn deall y mecansims sy'n gyrru patholegau cardiofasgwlaidd mewn cleifion â chlefyd cronig yr arennau (CKD). Mae CKD bellach yn ail yn nifer yr achosion o glefyd y galon yn y byd datblygedig. Mae gan gleifion sydd â CKD cyfnod terfynol sydd ar ddialysis ganlyniadau tebyg (os nad gwaeth) i lawer o ganserau, ond nid yw hyn yn cael ei gydnabod yn dda. Diasitis cardiofasgwlaidd yw'r prif achos o afiachusrwydd a marwolaeth yn y grŵp hwn o gleifion. Yn anffodus, nid ydynt yn ymateb yn dda i therapïau cardiofasgwlaidd traddodiadol fel therapi gwrth-blatennau, therapi gostwng lipidau neu angioplasti percutaneous. Nod fy grŵp yw nodi'r mecanweithiau sy'n gyrru patholeg cardiofasgwlaidd mewn cleifion â chlefyd arennol ac ymchwilio i dargedau therapiwtig newydd. Mae meysydd ymchwil cysylltiedig eraill o fewn neffroleg yn cynnwys deall y mecanweithiau sy'n gyrru ffibrosis peritoneol ac sy'n arwain at fethiant dialysis peritoneol (PD) mewn cleifion â chlefyd arennol ac rwy'n un o'r Ymchwilwyr Prifathrawiaethol yn rhaglen ITN IMPROVE PD Marie Curie a ariennir gan Horizon 2020.
Ymchwil COVID19: Rwy'n cydweithio â grŵp o fewn y Gyfarwyddiaeth Neffroleg a Thrawsblaniadau clinigol yn ogystal â grwpiau ymchwil Imiwnoleg a Virolgy ym Mhrifysgol Caerdydd i ymchwilio i ymatebion brechlyn COVID19 mewn cleifion ar haemodialysis, gydag astudiaethau tebyg yn cael eu harwain mewn cleifion tranplanhigyn arennau sy'n cael eu rhedeg gan yr un grŵp.

Fi yw'r Arweinydd Arbenigedd ar gyfer addysg feddygol israddedig mewn Meddygaeth Arennol yn y Gyfarwyddiaeth Neffroleg a Thrawsblannu ac rwy'n Gadeirydd pwyllgor Llywodraethu Addysg yr adran, sy'n goruchwylio gwelliant mewn addysg a hyfforddiant arennol israddedig ac ôl-raddedig. Rwy'n eistedd ar bwyllgor Arennol STC yng Nghymru ac rwy'n Oruchwyliwr Addysgol a Chlinigol i nifer o hyfforddeion arbenigol a hyfforddeion y Rhaglen Sylfaen.
Rwy'n goruchwylio prosiectau ymchwil ar gyfer myfyrwyr PhD ac yn croesawu ymholiadau i oruchwylio myfyrwyr newydd, gan gynnwys myfyrwyr Meistr, myfyrwyr meddygol Intercalated a myfyrwyr israddedig PTY.
Yn ddiweddar, dyfarnwyd Medal Medawar i'm myfyriwr PhD diweddar Dr Charlotte Brown yng nghyfarfod Cymdeithas Trawsblannu Prydain ym mis Chwefror 2021 am ymchwil a wnaed yn fy Labordy.
