Mae'r rhaglen ymchwil, sydd wedi'i lleoli yn yr adran Cemeg Fiolegol, mewn ecoleg gemegol sy'n cynnwys rhyngweithiadau cemegol rhwng organebau amrywiol gan gynnwys plâu sy'n ymosod ar blanhigion ac anifeiliaid. Ar ôl bod y cyntaf i nodweddu, fel strwythurau moleciwlaidd newydd gan gynnwys isoprenoidau, mae pheromones cysylltiedig rhyw fectorau pryfed, e.e. aphids, mosgitos a phryfed tywod, o bathogenau planhigion a dynol, nodweddu cemegol pheromones a semiochemicals eraill yn parhau i fod yn sail i lawer o'r ymchwil. Mae hyn yn cynnwys technolegau soffistigedig ar gyfer dal a thrafod meintiau munud y cemegau dan sylw a defnyddio cromatograffeg cydraniad uchel cypledig i sbectrometreg màs hynod sensitif gan gynnwys system Orbitrap Actif ThermoFisher ar gyfer adnabod petrusol. Ar hyn o bryd mae'r dull hwn yn cael ei gyfeirio at adnabod attractants ystod hir o fosgitos yn ystod y dydd gan ddefnyddio hefyd astudiaethau electroffisiolegol antena cypledig GC ar y lefel synhwyrol ffatri olfactory sengl gyda chydweithwyr yn Ysgol y Biowyddorau. Mae meysydd ymchwil cysylltiedig eraill:
- Er mwyn nodi, a manteisio ar reoli plâu, rydym yn astudio signalau cemegol sy'n gysylltiedig â straen o blanhigion, a fertebratau gan gynnwys anifeiliaid dynol a fferm, er enghraifft ar: (i) ymateb amddiffyn naturiol india-corn i blâu sy'n arwain at fwy o ymosodiad ar y plâu gan waspiaid parasitig, gyda chydweithwyr yn Affrica Is-Sahara; (ii) signalau cysylltiedig nad ydynt yn westeiwr yn erbyn arthropodau sy'n ymosod ar gamelod mewn hwsmonaeth anifeiliaid traddodiadol yn Somaliland; (iii) arwyddion cemegol ar gyfer diagnosis cynnar o ddatblygiad pathogenau ffwngaidd a firaol iechyd planhigion a dynol gyda gwahanol gydweithwyr yn y DU.
- Mae eucidating llwybrau biosynthetig i pheromones a semiochemicals eraill, ynghyd â geneteg moleciwlaidd cysylltiedig, yn cynnwys astudiaethau newydd ar fiosynthesis cyfansoddion sy'n denu plâu o blanhigion banana masnachol, ac o homosesquiterpenoids afreolaidd o pheromones rhyw hedfan tywod mewn cydweithrediad â Phrifysgol Nottingham.
- Mae manteisio ar pheromones a semiochemicals eraill wrth reoli plâu ac organebau buddiol trwy reoli ecoleg ac addasu genetig (GM) yn cynnwys astudiaethau cemegol sy'n sail i ddatblygu systemau gwthio-dynnu sy'n darparu amddiffyniad yn erbyn plâu (rehomoterpenes) a chwyn (re di-C-glycosylatedflavonoids) gyda gwahanol gydweithredwyr gan gynnwys yn Affrica Is-Sahara ac yn cynnwys asiantaethau masnachol wedi'u lleoli yn Ne Cymru.

Cydnabuwyd cyfraniadau John i faes ecoleg gemegol gyda Gwobr Rank 1995 am Faeth a Hwsmonaeth Cnydau, etholwyd ef i Gymrodoriaeth y Gymdeithas Frenhinol ym 1996, ethol i Aelodaeth y Deutsche Akademie der Naturforscher Leopoldina yn 2001 a derbyniodd Fedal Ryngwladol Cymdeithas Ecoleg Cemegol yn 2002. Penodwyd John i CBE am Wasanaethau i Gemeg Fiolegol yn 2004.
Yn 2008 dyfarnwyd Gwobr Wolf Foundation mewn Amaethyddiaeth iddo ar y cyd. Cyflwynodd brif ddarlith y Gymdeithas Frenhinol yn y gwyddorau biolegol, Darlith Gwobr Croonaidd yn 2008, a Darlith Prifysgol Cornell yn 2009. Dyfarnwyd iddo Dystysgrif Rhagoriaeth Cyngres Ryngwladol Entomoleg yng Nghyngres Ryngwladol Entomoleg XXIV a gynhaliwyd yng Nghorea, Awst 2012. Fe'i hetholwyd yn Aelod Rhyngwladol Academi Genedlaethol y Gwyddorau yn 2014. Daeth yn Llywydd y Gymdeithas Entomolegol Frenhinol yn 2014 ac ym mis Awst 2017 rhoddodd Ddarlith Gwobr Goffa Sterling B Hendricks yn 254ain Cyfarfod Cymdeithas Cemegol America, Washington DC. Yn 2020, etholwyd John yn Gymrawd Cymdeithas Ddysgedig Cymru.
