Mae'r ymennydd yn gofyn am gyflenwad cyson o egni, lle mae angen coreograffu llif y gwaed trwy rwydwaith cymhleth o bibellau gwaed i ateb gofynion ynni meinwe. Mae fy ymchwil wedi datgelu bod unigolion ifanc sydd â mwy o risg genetig ar gyfer clefyd Alzheimer (AD) wedi lleihau llif gwaed yr ymennydd yn sylweddol sy'n parhau i fod yn gyson ar draws y rhychwant oes, lle mae mynegiant genynnau risg AD rhanbarthol uwch yn gysylltiedig â llai o lif gwaed yr ymennydd. Mae fy ymchwil ynghyd ag eraill ym maes dementia wedi dangos bod dadansoddiad yn rheoleiddio system fasgwlaidd yr ymennydd yn un o'r newidiadau cynharaf y gellir eu canfod yng nghlwy'r clefyd Alzheimer. Felly, gall targedu system fasgwlaidd yr ymennydd ar gyfer strategaethau ataliol helpu i liniaru dysregulation fasgwlaidd yn y rhai sydd mewn perygl, ac oedi dilyniant pathoffisioleg sy'n gysylltiedig ag AD.
Rwyf wedi cefnogi dilysu dulliau MRI newydd sydd â chyfleustodau clinigol ar gyfer diagnosio dysregulation fasgwlaidd mewn clefydau. Rwyf bellach yn cymhwyso'r dulliau delweddu MRI newydd hyn i nifer o garfannau clinigol gan gynnwys sglerosis ymledol, dementia, epilepsi, ac arthritis gwynegol. Mae fy ngwaith hefyd yn cynnwys defnyddio methodolegau delweddu PET i asesu ffisioleg yr ymennydd. Fel ECR mewn delweddu PET, rwyf wedi ymuno â Dr Mattia Veronese a'r Athro Federico Turkheimer yng Ngholeg y Brenin, Llundain, lle gwnaethom gyd-sefydlu Partneriaeth PET y DU (PPUK). Datblygwyd y PPUK i alluogi cydweithrediadau ar raddfa fwy ledled y DU a thu hwnt i fynd i'r afael â chwestiynau ymchwil pwysig wrth astudio clefydau. Mae hefyd yn darparu adnodd i ECR ddod o hyd i gymorth gyda dylunio astudiaeth ddelweddu PET, casglu a dadansoddi data.
