Sut mae celloedd mamaliaid yn rheoleiddio ac yn cydlynu'n ofodol y broses o symud cargo cellog, tra'n sicrhau bod homeostasis organelle, yn cael ei gynnal a chargos yn cael eu cyflwyno'n gywir, yw ffocws fy ymchwil. Gan ddefnyddio atalyddion moleciwlau bach, tocsinau, a bioleg celloedd clasurol, a defnyddio dulliau optegol fflworoleuedd ac aflinol, rwy'n astudio'r llwybrau y mae celloedd mamaliaid yn gallu symud proteinau, lipidau a nanoronynnau rhwng adrannau cellog.

Microsgopeg CEIR
Mewn cydweithrediad â Dr Paola Borri a'r Athro Wolfgang Langbein, rydym yn defnyddio microsgopeg gwasgariad cylynol gwrth-stokes Raman i astudio homeostasis lipid o fewn celloedd ewcaryotig. Mae microsgopeg CARS yn ymuno â'r sensitifrwydd cemegol a noninvasiveness di-label a gynigir gan sbectrosgopeg dirgrynol gyda'r gallu adrannol 3D cynhenid microsgopeg multiphoton. Yn ddiweddar, rydym wedi cyhoeddi ein gwaith ar ddull o berfformio CEIR GWAHANIAETHOL AMLEDD (D-CARS) mewn Llythyrau Opteg.
Delweddu cymysgu Pedair Ton (FWM)
Mewn cydweithrediad â Dr Paola Borri a'r Athro Wolfgang Langbein, rydym wedi dangos microsgop amlffoton newydd yn seiliedig ar ganfod pedair ton yn cymysgu a allyrrir o nanoronynnau aur. Mae hyn yn caniatáu inni berfformio delweddu rhydd cefndir o labeli aur sengl o feintiau bach (hyd at 5nm) gyda datrysiad is-micromedr. Rydym wedi cyhoeddi'r gwaith hwn yn ddiweddar yn Optics Letters ac rydym bellach yn ymchwilio i gymhwysedd y dechneg hon i'r gwyddorau bywyd.

Mae gennym nifer o ficrosgopau yn y labordy sydd ar gael i'w defnyddio:
- CRAM: Widefield timelapse cyflym ffurfweddu ar gyfer delweddu fflworoleuedd DI / fitc / tritc / Cy5 a chyferbyniad cam ar gyfer delweddu brightfield.
- Bernard: Widefield timelapse cyflym wedi'i ffurfweddu ar gyfer delweddu fflworoleuedd dapi / fitc / tritc / Cy5 a chyferbyniad cam ar gyfer delweddu brightfield. Microsgop disg nyddu ar gyfer DAPI / FITC / TRITC
Mae gennym hefyd system microbigiad eppendorf a fydd yn cyd-fynd â phob un o'r microsgopau hyn i ganiatáu microbigiad, neu ficro-ddosbarthu deunydd i'r gell, neu ei hamgylchedd lleol.

Enillais fy ngradd gyntaf mewn Biocemeg Feddygol o Brifysgol Birmingham, a chefais fy PhD dan oruchwyliaeth yr Athro John Davey ym Mhrifysgol Warwick, gan astudio llwybr cyfathrebu pheromone burum ymhollt. Gan aros ym Mhrifysgol Warwick, yn 2000 dechreuais astudio masnachu tocsin yn labordai yr Athro Mike Lord a Lynne Roberts, ac yn ystod y cyfnod hwn roeddwn yn ddigon ffodus i gael fy hyfforddi mewn microsgopeg ysgafn gan Dr. Jez Simpson a Dr. Rainer Pepperkok yn EMBL yn Heidelberg.
Tua diwedd 2003 symudais i labordy Dr. David Stephens ym Mhrifysgol Bryste, lle defnyddiais ficrosgopeg golau ac electron i edrych ar sut mae proteinau yn gadael yr ER ac yn cael eu symud trwy'r gell mewn cludwyr trafnidiaeth membranaidd.
Ym mis Chwefror 2007 symudais i Ysgol y Biowyddorau ym Mhrifysgol Caerdydd i dderbyn Cymrodoriaeth RCUK mewn Ymchwil Drosiadol mewn Meddygaeth Arbrofol.
